The first major step in the cycle of eukaryotic transcription - INITIATION! We talk about the assembly of general transcription factors (GTFs) associated with RNA polymerase II and look at the role of each GTF in the mechanistic production of a pre-initiation complex (PIC). The PIC formation is linked to the enhancer and we also see how they work together. After the formation of pre-initiation complex, the RNA polymerase II needs to clear the promoter region but when it does it pauses again - we discuss why that occurs.
